Qualitative Exclusion of Iodide in 5-Amino-1MQ Chloride (CAS 42464-96-0) via Starch-Peroxide Oxidation

1. Background and Chemical Identity

CAS number 42464-96-0 is frequently, and erroneously, cross-listed for both 5-Amino-1-methylquinolinium chloride and 5-Amino-1-methylquinolinium iodide. Albochemicals LLC explicitly supplies the Chloride salt. To ensure material integrity prior to formulation, researchers can perform a simple qualitative limit test to verify the complete absence of the iodide counterion.

2. Reagents and Protocol

This assay relies on the oxidation of iodide ions (I–) into elemental iodine (I2) by a strong oxidizing agent, which subsequently reacts with amylose (starch) to form a highly visible coordination complex.

Sample: 50 mg of 5-Amino-1MQ Chloride test material.

  • Solvent: 5 mL of distilled water (H2O).
  • Oxidizing Agent: 1 mL of 3% Hydrogen Peroxide (H2O2).
  • Indicator: 0.5 mL of standard starch indicator solution.

Dissolve the test material completely in the distilled water. Add the starch indicator solution and agitate gently. Introduce the hydrogen peroxide dropwise. Observe the solution against a white background for 60 seconds.

3. Result Interpretation

  • Negative Result (Pass): The solution remains clear or retains the natural pale tint of the dissolved API. No  black coloration occurs. This confirms the exclusion of the iodide counterion and verifies the material as the Chloride salt.
  • Positive Result (Fail): The immediate formation of an opaque, dark blue-black complex. This indicates the presence of elemental iodine, confirming that the sample is the Iodide salt or is cross-contaminated with other Iodine containing products.
